Nestin Expressing Progenitors

Nestin-expressing progenitors are immature neural cells identified by expression of nestin, an intermediate filament protein associated with developing and stem-like cells in the nervous system. Nestin expression is typically high in undifferentiated progenitors and decreases as they differentiate into neurons, astrocytes, or oligodendrocytes, making it a useful marker of changing cell state. In neuroscience, researchers use nestin immunostaining, reporter systems, and lineage-tracing approaches to locate and characterize neural progenitor populations. Studying these cells helps clarify brain development, adult neurogenesis, neural injury responses, and the cellular basis of strategies for nervous-system repair.

A Quantitative Assay for Insulin-expressing Colony-forming Progenitors

A three-dimensional clonogenic assay that allows pancreatic-like progenitors to differentiate into insulin-expressing colonies is described. This method takes advantage of semi-solid media containing methylcellulose, Matrigel and growth factors, in which single progenitors proliferate and differentiate in vitro, permitting quantification of the number of functional progenitors in a population.

Marrow stromal cells (MSCs) with neural potential exist within the bone marrow. Our protocol enriches this population of cells via hypoxic preconditioning and thereafter directs them to become mature Schwann cells.

Here we describe an optimized, highly reproducible protocol to isolate Mesodermal Progenitor Cells (MPCs) from human bone marrow (hBM). MPCs were characterized by flow cytometry and nestin expression. They showed the ability to give rise to exponentially growing MSC-like cell cultures while retaining their angiogenic potential.
