Synaptophysin Colocalization

Synaptophysin colocalization is an imaging-based method for identifying and characterizing synaptic vesicle sites by assessing where synaptophysin overlaps with other cellular markers. Synaptophysin, an abundant membrane protein of synaptic vesicles, is detected through immunofluorescence or fluorescent tagging, while microscopy and image-analysis methods measure signal overlap with proteins such as presynaptic, postsynaptic, or neuronal markers. In neuroscience, this approach helps map synaptic distribution, evaluate neuronal connectivity, and compare changes in synapse organization across development, disease models, or experimental treatments. Quantitative colocalization can provide evidence of spatial association, although it should be interpreted alongside complementary structural or functional assays.

2026

Source: Zhang, L., et al. Immunofluorescent Labeling of Plant Virus and Insect Vector Proteins in Hemipteran Guts. J. Vis. Exp. (2021)The video demonstrates the preparation of gut tissue from a plant virus–infected insect vector for fluorescence imaging. Fixed guts were washed, permeabilized, and incubated with two fluorescent antibodies in a blocking solution—one targeting a viral antigen in vesicles, the other a vesicle-associated membrane protein. The tissue was counterstained for actin...

Visualizing individual cells in densely packed tissues, such as terminal Schwann cells (SCs) at neuromuscular junctions (NMJs), is challenging. "Sequential photo-bleaching" allows delineating single terminal SCs, for instance in the triangularis sterni muscle explant, a convenient nerve-muscle preparation, where sequential bleaching can be combined with time-lapse imaging and post-hoc immunostainings.
