Neural Stem Cell Culture

Neural stem cell culture is the laboratory maintenance and expansion of self-renewing, multipotent cells that can generate neurons, astrocytes, and oligodendrocytes, providing a controlled model for neuroscience research. Researchers typically isolate neural stem cells from neural tissue or stem cell-derived populations and grow them in defined, often serum-free media containing mitogens such as epidermal growth factor and fibroblast growth factor 2; removing these signals can promote differentiation. Cultured cells support studies of neural development, disease mechanisms, neurotoxicity, and regenerative strategies. Their behavior can be assessed through neurosphere formation, proliferation measurements, and lineage-specific markers, helping researchers evaluate how genes, drugs, or environmental conditions influence nervous system cells.

Here is a protocol to grow pluripotent stem cells (PSC) and neural stem cells (NSC) in an enclosed cell culture system that permits maximum sterility and reproducibility, replacing the traditional biosafety cabinet and incubator. This equipment meets clinical good manufacturing practice (cGMP) and clinical good lab practice (cGLP) guidelines.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.

Isolation and Culture of Adult Neural Stem Cells from the Mouse Subcallosal Zone

Establishing culture systems for the expansion of adult neural stem cells (aNSCs) allows for the examination and application of aNSCs for therapy. The subcallosal zone (SCZ) has recently been recognized as a novel neuroblast-forming region in adult mice. Here, methods for the isolation, expansion, and differentiation of SCZ-aNSCs are described.
