Stable Gene Manipulation

Stable gene manipulation is the deliberate alteration of an organism’s genetic material so that the introduced or modified sequence persists over time and can be maintained in descendant cells. It works by editing a target locus or integrating an engineered DNA construct into the genome, then identifying and validating cells that retain the intended change through selection, genotyping, or expression analysis. In neuroscience, stable manipulation supports the study of gene function, neural development, circuit activity, and disease mechanisms by producing consistent cellular or animal models; it can also enable long-term expression of reporters or therapeutic genes for evaluating neuronal phenotypes and interventions.

We describe approaches for the manipulation of genes in the evolutionary model system Astyanax mexicanus. Three different techniques are described: Tol2-mediated transgenesis, targeted manipulation of the genome using CRISPR/Cas9, and knockdown of expression using morpholinos. These tools should facilitate the direct investigation of genes underlying the variation between surface- and cave-dwelling forms.
