Submucosal Pocket

A submucosal pocket is a surgically created space beneath a mucosal lining that provides a protected site for placing biological materials, sensors, or implanted devices. It is formed by gently separating the mucosa from the underlying connective tissue, creating a tissue plane that can accommodate the selected material while limiting direct exposure to the external environment. In neuroscience, this approach can support the placement of neural interfaces or delivery systems near sensory or peripheral neural structures, helping investigators study neural signaling and evaluate device compatibility. Its use also highlights how anatomy, tissue healing, and implant stability influence experimental outcomes.

The cornea is unique in that it lacks vascular tissues. However, robust blood vessel growth and survival can be induced in the cornea by potent angiogenic factors. Therefore, the cornea can provide with us a valuable tool for angiogenic studies. This protocol demonstrates how to perform the mouse model of cornea pocket assay and how to assess the angiogenesis induced by angiogenic factors using this model.

Here we demonstrate our protocol for isolation of basal and submucosal gland duct cells from mouse tracheas. We also demonstrate the method of injecting stem cells into the dorsal mouse fat pad to create an in vivo model of submucosal gland regeneration.
