Synaptic Connection Analysis

Synaptic connection analysis is the study of how neurons communicate at synapses and how their connections are organized, strengthened, or weakened. Researchers assess synaptic structure and function by combining approaches such as neural tracing, microscopy, electrophysiology, and measurement of synaptic transmission, including changes in postsynaptic electrical responses. In neuroscience, these analyses reveal how neural circuits encode information, coordinate behavior, and adapt through plasticity. They also help characterize circuit disruption in neurological and psychiatric disorders, evaluate the effects of drugs or injury, and guide research on learning, memory, development, and potential strategies for restoring impaired connectivity.

Studying Synaptic Vesicle Pools using Photoconversion of Styryl Dyes

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.

This paper describes the procedures for tactile stimulation of rat pups and subsequent Golgi-Cox staining of neuronal morphology. Tactile stimulation is a positive experience that is administered in the perinatal period by stroking pups with a household duster. Golgi-cox staining is a reliable procedure permitting the visualization of entire neurons.

Automated Quantification of Synaptic Fluorescence in C. elegans

The abundance of neurotransmitter receptors clustered at synapses strongly influences synaptic strength. This method quantifies fluorescently-labeled neurotransmitter receptors in three dimensions with single-synapse resolution in C. elegans, allowing hundreds of synapses to be rapidly characterized within a single sample without distortions introduced by z-plane projection.
