Respirometric Assay

A respirometric assay is a method for measuring biological respiration by tracking oxygen consumption, carbon dioxide production, or changes in pressure over time. In biochemistry, the assay places cells, microorganisms, mitochondria, or enzymes in a controlled chamber and detects gas exchange as substrates are oxidized, linking respiratory activity to energy metabolism. Researchers use respirometric assays to evaluate metabolic rates, mitochondrial function, microbial growth, substrate utilization, and responses to inhibitors or environmental conditions. Because measurements are quantitative and time-resolved, this approach helps characterize cellular bioenergetics and assess how biochemical pathways respond to experimental changes.

Here, we describe a detailed method for mitochondria isolation from mouse skeletal muscle and the subsequent analysis of respiration by Oxygen Consumption Rate (OCR) using microplate-based respirometric assays. This pipeline can be applied to study the effects of multiple environmental or genetic interventions on mitochondrial metabolism.
