High Performance Microscopy

High performance microscopy is the use of advanced optical systems, detectors, and computational methods to capture biological structures and processes with high spatial or temporal resolution. These systems combine optimized illumination, sensitive image detection, precise sample control, and quantitative image analysis to resolve dynamic events while limiting photodamage and other imaging artifacts. In biological techniques, high performance microscopy supports the study of cell organization, molecular interactions, tissue architecture, and time-dependent biological processes. Its measurements can reveal mechanisms that conventional imaging may miss and provide quantitative data for cell biology, developmental studies, disease research, and drug evaluation.

Using a Laminating Technique to Perform Confocal Microscopy of the Human Sclera

Human sclera tissue is mainly collagen; therefore, it is not easily usable for immunohistochemistry. To achieve the goal of performing immunohistochemistry for confocal microscopy of scleral tissue, a laminating technique was used.

Taking advantage of intravital microscopy, the method presented here enables real-time visualization of intestinal epithelial cell shedding in living animals. Therefore, topically stained intestinal mucosa (acriflavine and rhodamineB-dextran) of anesthetized mice is imaged up to single-cell resolution using confocal microscopy.

A method is described whereby quantum dot (QD) nanoparticles can be used for correlative immunocytochemical studies of epoxy embedded human pathology tissue. We employ commercial antibody fragment conjugated QDs that are visualized by widefield fluorescence light microscopy and transmission electron microscopy.
