Embryo Dissociation

Embryo dissociation is a laboratory technique that separates an early embryo into individual cells or small cell clusters for analysis and culture. It typically combines enzymatic digestion, which weakens extracellular matrix and cell-adhesion proteins, with controlled mechanical pipetting to disrupt tissue while preserving cell viability. The resulting suspension supports cell counting, lineage characterization, single-cell molecular analysis, and establishment of cultures or organoid models. In developmental biology, embryo dissociation helps researchers examine how cells acquire distinct identities, respond to signaling environments, and contribute to tissue formation, providing a foundation for studying development, disease mechanisms, and regenerative applications.

We describe a rapid, inexpensive, and reliable method to isolate, dissociate, and culture sensory neurons from chick embryos. Details of substrata preparation and immunocytochemistry are also provided.

Zebrafish are an emerging system for modeling human disorders of the skeletal muscle. We describe a fast and efficient method to isolate skeletal muscle myofibers from embryonic and larval zebrafish. This method yields a high-density myofiber preparation suitable for study of single skeletal muscle fiber morphology, protein subcellular localization, and muscle physiology.
