Genetic Tractable Microbes

Genetically tractable microbes are microorganisms that can be reliably grown, modified, and analyzed using laboratory genetic tools, making them valuable systems for biology research. Their tractability typically depends on efficient DNA delivery, selectable markers, and methods such as homologous recombination or CRISPR-based editing, which enable targeted changes to genes and their evaluation under defined conditions. Bacteria, yeasts, and other tractable microbes support studies of gene function, regulation, metabolism, host-microbe interactions, and evolution, while also serving as platforms for biotechnology. These organisms help link genotype to phenotype and accelerate the development of engineered pathways, useful products, and experimental models.

Genetic Manipulation of the Plant Pathogen Ustilago maydis to Study Fungal Biology and Plant Microbe Interactions

We describe a robust gene replacement strategy to genetically manipulate the smut fungus Ustilago maydis. This protocol explains how to generate deletion mutants to investigate infection phenotypes. It can be extended to modify genes in any desired way, e.g., by adding a sequence encoding a fluorescent protein tag.

Tractable Mammalian Cell Infections with Protozoan-primed Bacteria

This technique provides a method to harvest, normalize and quantify intracellular growth of bacterial pathogens that are pre-cultivated in natural protozoan host cells prior to infections of mammalian cells. This method can be modified to accommodate a wide variety of host cells for the priming stage as well as target cell types.

Extracting DNA from the Gut Microbes of the Termite (Zootermopsis Angusticollis) and Visualizing Gut Microbes

This video illustrates the technique for extracting DNA from the species of microbes resident in the termite hindgut. The preparation of a wet mount slide, which is useful for visualizing the gut microbial community is also illustrated, and a tour through the species-rich gut environment is given.
