Two Chamber Assay

A two-chamber assay is an in vitro experimental system that separates biological samples or cell populations into two compartments, allowing researchers to study interactions across a controlled boundary. Typically, the chambers are divided by a porous membrane that permits soluble factors, nutrients, or migrating cells to pass between compartments while keeping the main populations physically distinct. In biology, this design supports measurements of cell migration, invasion, chemotaxis, permeability, paracrine signaling, and cell-cell communication. By varying membrane properties, chemical gradients, or culture conditions, researchers can isolate mechanisms and evaluate how cells respond to neighboring tissues, signaling molecules, drugs, or other experimental treatments.

Human Neutrophil Flow Chamber Adhesion Assay

A method of quantitating neutrophil adhesion is reported. This method creates a dynamic flow environment similar to that encountered in a blood vessel. It allows the investigation of neutrophil adhesion to either purified adhesion molecules (ligand) or endothelial cell substrate (HUVEC) in a context similar to the in vivo environment with sheer stress.

Analysis of Trunk Neural Crest Cell Migration using a Modified Zigmond Chamber Assay

An approach to analyze the migration of explanted cells (trunk neural crest cells) is described. This method is inexpensive, gentle, and capable of distinguishing chemotaxis from both chemokinesis and other influences on migratory polarity such as those derived from cell-cell interactions within the primary trunk neural crest cell culture.
