Cell Recellularization

Cell recellularization is the process of repopulating a decellularized tissue or organ scaffold with living cells, restoring cellular components within an extracellular matrix framework. Researchers remove native cells while retaining matrix architecture and biochemical cues, then introduce selected cells through seeding or perfusion-based culture under conditions that support attachment, survival, and distribution. In cancer research, recellularized scaffolds can incorporate tumor and stromal cells to model tumor microenvironments, investigate invasion and cell–matrix interactions, and evaluate therapeutic responses. These three-dimensional systems can improve the physiological relevance of preclinical studies and support research into personalized treatment strategies.

Decellularization and Recellularization of Whole Livers

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.

The description of a nonhuman primate model of lung regeneration in which whole lungs are decellularized and then seeded with adult stem cells and endothelial cells in a bioreactor that facilitates vascular circulation and liquid media ventilation is presented.
