Primary Cell Isolation

Primary cell isolation is the process of obtaining viable, directly derived cells from tissues or biological samples for laboratory study, preserving characteristics that may be altered in long-term cell lines. In cancer research, tissue is dissociated through mechanical disruption, enzymatic digestion, or both, followed by separation and washing steps that enrich the desired cell population while maintaining cell viability and function. Isolated tumor, stromal, or immune cells can support ex vivo culture, molecular profiling, drug screening, and investigation of tumor–microenvironment interactions. Because these cells retain patient- or tissue-specific features, primary cell isolation helps connect experimental findings with disease biology and personalized research.

This protocol describes a new technique that completely inverts blood vessels of interest, exposing only the endothelial side to enzymatic digestion.
