Ifn-gamma Secretion Detection

IFN-γ secretion detection is the measurement of interferon-gamma released by activated immune cells, providing a functional readout of cell-mediated immune responses during infection and inflammation. It typically involves stimulating lymphocytes with an antigen or mitogen, then capturing or labeling secreted cytokine with specific antibodies for quantification by ELISA, ELISpot, or intracellular cytokine staining. These assays can reveal responding cell frequencies, secretion levels, and antigen-specific activity while distinguishing functional immune responses from simple cell presence. In immunology and infection research, the results support analysis of host-pathogen interactions, immune competence, vaccine responses, and disease-associated inflammation, helping investigators compare conditions and evaluate immune protection.

This video demonstrates an imaging method to visualize in situ interferon-gamma (IFN-γ) secretion in the mouse spleen. Following bacterial infection and the induction of IFN-γ accumulation inside T cells and natural killer cells, the spleen is harvested from the mouse. Upon obtaining tissue sections, immunostaining is performed to visualize IFN-γ inside the cells.
