Nss Gene Manipulation

Nss gene manipulation is the targeted alteration, deletion, or replacement of an Nss gene to determine how its encoded product influences pathogen biology and host responses. Using molecular cloning, site-directed mutagenesis, or reverse-genetics systems, researchers generate defined gene variants and compare their replication, protein expression, and interactions with infected cells against an unmodified control. In immunology and infection research, these approaches help identify Nss-dependent effects on innate immune signaling, immune evasion, and pathogenicity. The resulting mutants can clarify virulence mechanisms, reveal potential therapeutic targets, and support the development of attenuated strains or improved experimental models.

The reverse genetics system for the Rift Valley fever virus MP-12 vaccine strain is a useful tool for creating additional MP-12 mutants with increased attenuation and immunogenicity. We describe the protocol to generate and characterize NSs mutant strains.

We describe approaches for the manipulation of genes in the evolutionary model system Astyanax mexicanus. Three different techniques are described: Tol2-mediated transgenesis, targeted manipulation of the genome using CRISPR/Cas9, and knockdown of expression using morpholinos. These tools should facilitate the direct investigation of genes underlying the variation between surface- and cave-dwelling forms.
