Cell Transduction Protocol

A cell transduction protocol is a stepwise method for delivering genetic material into cells, usually with a viral vector, to alter cellular function for medical research or treatment. The vector binds cellular receptors, enters the cell, and releases its nucleic acid cargo, which may be expressed temporarily or integrated into the host genome depending on the vector system. Researchers optimize cell type, vector dose, exposure time, and culture conditions to achieve efficient delivery while limiting toxicity. These protocols support gene therapy development, engineered immune-cell therapies, disease modeling, and studies of gene function, making them important tools in modern medicine.

Lentiviruses are a valuable research tool for exploring gene function; however, researchers may wish to avoid production of pantropic lentivirus encoding known or suspected oncogenes. The goal of this study was to formulate technologies that allow for successful gene transduction in primary natural killer (NK) cells. The dextran-mediated lentiviral transduction of human or mouse primary NK cells results in higher gene expression efficiencies. This method of gene transduction will vastly improve NK cell genetic manipulation.
