Intracerebroventricular Injection

Intracerebroventricular injection is a technique that delivers drugs, biological agents, or experimental compounds directly into the brain’s ventricular system, where cerebrospinal fluid circulates. A needle or catheter is positioned within a cerebral ventricle, allowing the injected substance to disperse through cerebrospinal fluid and partly bypass the blood-brain barrier. In medicine and biomedical research, this approach supports studies of central nervous system pharmacology, neurological disease mechanisms, and treatments that cannot readily reach brain tissue through systemic administration. Its effectiveness depends on accurate anatomical placement, controlled dosing, and careful assessment of potential tissue injury, altered intracranial pressure, or other procedure-related effects.

The amyloid-β (Aβ)-injected animal model enables the administration of a defined quantity and species of Aβ fragments and reduces individual differences within each study group. This protocol describes the intracerebroventricular (ICV) injection of Aβ without stereotactic instruments, enabling the production of Alzheimer-like behavioral abnormalities in normal mice.
