Dendrite Harvesting

Dendrite harvesting is a neuroscience technique for selectively collecting dendritic compartments from neurons so their molecular composition can be studied separately from cell bodies and axons. In cultured neurons, researchers isolate or physically remove dendrites under controlled conditions, then preserve the harvested material for downstream analysis of RNA, proteins, or other cellular components. This compartment-specific approach helps investigate local gene expression, protein synthesis, synaptic signaling, and structural changes associated with neuronal development and plasticity. By linking dendritic molecules to neuronal function, dendrite harvesting supports research into circuit formation, learning and memory, and neurological disease mechanisms.

Selective Harvesting of Marginating-pulmonary Leukocytes

Marginating-pulmonary leukocytes exhibit unique characteristics and distinct immunological functions compared to other leukocyte populations. Here we describe selective harvesting of this subpopulation of pulmonary leukocytes, by forced perfusion of the lungs in rats and mice. Marginating-pulmonary leukocytes seem critical in determining susceptibility to blood-borne and lung-related diseases.

Preparation of Tumor Antigen-loaded Mature Dendritic Cells for Immunotherapy

The most commonly used method for generating large numbers of autologous dendritic cells (DCs) for use in tumor immunotherapy is described. The method uses IL-4 and GM-CSF to differentiate DCs from monocytes. The immature DCs are stimulated to mature and then loaded with antigens before they are injected back into the patient.

This video demonstrates the procedure for differentiating myeloid dendritic cells from mouse bone marrow. Isolation of mouse tibia and femur, and processing of bone marrow are demonstrated. Pictures demonstrating cell morphology before and after differentiation, and figures depicting cell phenotype and IL-12 production following maturation using CpG are shown.
