Double Diagonal Brain Cutting

Double diagonal brain cutting is a neuroanatomical dissection technique that uses two angled cuts to divide brain tissue into defined blocks for examination and analysis. By orienting the cuts relative to recognizable anatomical landmarks, researchers can expose targeted regions while preserving spatial relationships and producing consistent tissue faces for subsequent sectioning, staining, or imaging. The method supports accurate localization of structures in neuroscience studies, including microscopic analysis, lesion assessment, and preparation of specimens for histology. Standardized cutting improves reproducibility across samples and helps connect observed cellular features with their anatomical position, strengthening interpretation of brain organization and experimental outcomes.

Organized brain cutting procedures are necessary to correlate specific neuropsychiatric phenomena with definitive neuropathologic diagnoses. Brain cuttings are performed differently based on various clinico-academic contingencies. This protocol describes a symmetric bihemispheric brain cutting procedure to investigate hemispheric differences in human brain pathologies and to maximize current and future biomolecular/neuroimaging techniques.

Double-barreled and Concentric Microelectrodes for Measurement of Extracellular Ion Signals in Brain Tissue

We demonstrate the fabrication, calibration and properties of two types of ion-selective microelectrodes (double-barreled and concentric) for measurement of ion concentrations in brain tissue. These are then used in the mouse hippocampal slice preparation to show that excitatory activity changes both extracellular potassium and sodium concentrations.

The video demonstrates the preparation of colliculo-thalamocortical brain slices from mouse pup brains. An isolated brain is aligned on a marked slide and cut at an angle to align the auditory midbrain and forebrain in the same plane. The brain is then sliced using a vibratome, and the slices containing the major auditory midbrain and forebrain structures are transferred to a holding chamber for preservation.
