Intracerebroventricular Infusion

Intracerebroventricular infusion is a method of delivering drugs or other experimental substances directly into the brain’s ventricular system, where they enter cerebrospinal fluid and can act on the central nervous system. A catheter or needle introduces the solution into a cerebral ventricle, while an infusion pump can regulate the dose and delivery rate, helping bypass much of the blood-brain barrier. In pharmacology, this technique supports studies of central drug action, dose-response relationships, pharmacokinetics, and neurobehavioral effects. Accurate placement and controlled administration are essential because imprecise delivery can alter experimental outcomes or damage tissue.

The amyloid-β (Aβ)-injected animal model enables the administration of a defined quantity and species of Aβ fragments and reduces individual differences within each study group. This protocol describes the intracerebroventricular (ICV) injection of Aβ without stereotactic instruments, enabling the production of Alzheimer-like behavioral abnormalities in normal mice.

To study the effects of Aβo in vivo, we developed a model based on repeated hippocampal infusions of soluble Aβo coupled with continuous infusion of Aβo antibody (6E10) in the hippocampus using osmotic pumps to counteract the neurotoxic effect of Aβo.
