Global Fitting Analysis

Global fitting analysis is a quantitative method that fits multiple experimental datasets simultaneously to a shared mathematical model, allowing researchers to identify parameters that describe related measurements as a whole. In biochemistry, the approach uses nonlinear regression to link datasets through common parameters while permitting experiment-specific variables, such as signal amplitude or baseline, to vary. By evaluating all measurements together, global fitting can improve parameter estimation, test mechanistic models, and reveal relationships that separate analyses may miss. It is commonly applied to enzyme kinetics, binding studies, spectroscopy, and reaction-rate analysis, supporting more rigorous interpretation of biochemical mechanisms and experimental variability.

QFA estimates the effect of genetic mutations, drugs or other applied treatments on microbe growth. Analysis of Global RNA Synthesis at the Single Cell Level following Hypoxia

We describe a technique for analysis of global RNA synthesis in hypoxia using imaging. Click-chemistry labeling of RNA has not previously been performed under hypoxia and allows visualization of global RNA changes at the single cell level. This approach complements the existing averaged RNA techniques, allowing direct visualization of cell-to-cell changes in global RNA synthesis.

Accurate, causality-based quantification of global diastolic function has been achieved by kinematic modeling-based analysis of transmitral flow via the Parametrized Diastolic Filling (PDF) formalism. PDF generates unique stiffness, relaxation, and load parameters and elucidates 'new' physiology while providing sensitive and specific indexes of dysfunction.
