Lipid Vesicle Model

Lipid vesicle model is a simplified experimental representation of a cell membrane, consisting of a closed lipid bilayer that encloses an aqueous compartment. In water, amphipathic phospholipids spontaneously self-assemble with hydrophobic tails shielded inside and hydrophilic heads facing the surrounding solution and internal contents, producing a selectively permeable boundary. These vesicles provide a controllable system for examining membrane organization, molecular transport, permeability, fusion, and interactions with proteins or external compounds, while supporting studies of cellular compartmentalization and processes such as drug delivery and synthetic biology.

Lipid Bilayer Vesicle Generation Using Microfluidic Jetting

Microfluidic jetting against a droplet interface lipid bilayer provides a reliable way to generate vesicles with control over membrane asymmetry, incorporation of transmembrane proteins, and encapsulation of material. This technique can be applied to study a variety of biological systems where compartmentalized biomolecules are desired.

Constant Pressure-controlled Extrusion Method for the Preparation of Nano-sized Lipid Vesicles

This protocol describes an extrusion method for preparing lipid vesicles of sub-micron sizes with a high degree of homogeneity. This method uses a pressure-controlled system with controlled nitrogen flow rates for liposome preparation. The lipid preparation1,2, liposome extrusion, and size characterization will be presented herein.

To test the interaction of a protein with its target lipid we used MACS and Annexin V-conjugated magnetic beads and lipid vesicles synthesized from the target lipid and Annexin V-binding phosphatidylserine. Proteins bound to the target lipid are co-purified and analyzed after elution from the beads.

This article presents a method for preparing compound Giant Unilamellar Vesicles (cGUVs) with a vesicle-in-vesicle structure, with customized electrical conductivity in inner, annular, and outer regions. Electroformation synthesizes simple GUVs, which are transformed into stomatocytes and cGUVs via osmotic shock, providing a valuable model for studying the biophysics of nucleated cells.
