Germline Toxicology Model

A Germline Toxicology Model is an experimental system for assessing how chemical, environmental, or physical exposures affect germ cells and may influence reproductive or offspring outcomes. It works by exposing developing germ cells, gonads, or suitable model organisms under controlled conditions, then examining endpoints such as cell survival, DNA damage, chromosome abnormalities, epigenetic changes, and altered developmental potential. In developmental biology, these models help connect toxicant exposure with germ-cell formation, maturation, and early embryonic development, supporting hazard identification, reproductive-risk assessment, and investigation of effects that may persist across generations.

We describe the detailed steps of a high-throughput chemical assay in the nematode Caenorhabditis elegans used to assess germline toxicity. In this assay, disruption of germline function following chemical exposure is monitored using a fluorescent reporter specific to aneuploid embryos.

We present an immunofluorescence imaging-based method for spatial and temporal localization of active ERK in the dissected C. elegans gonad. The protocol described here can be adapted for visualization of any signaling or structural protein in the C. elegans gonad, provided a suitable antibody reagent is available.

The objective of the study was to assess the biological impact of 15 cigarette smoke constituents using a combination of an impedance-based real time cell analyzer and a high-content screening (HCS)-based platform for toxicological assessment in vitro. This study provides information on effective doses, toxicity and modes of action of the tested compounds.
