Lineage Commitment

Lineage commitment is the biological process by which a progenitor or stem cell progressively restricts its developmental potential toward a defined cell fate, making it central to how multicellular organisms form specialized tissues. During development, extracellular signals activate lineage-specific transcription factors and epigenetic programs that stabilize gene expression while suppressing alternative fates. Studying lineage commitment helps researchers map cell-fate decisions, identify transitional progenitors, and understand how disruptions contribute to developmental disorders; the concept also guides stem-cell differentiation strategies for regenerative medicine and disease modeling.

A method to analyse the distribution of bone marrow hematopoietic progenitors in flow cytometry as well as to efficiently isolate highly purified hematopoietic stem cells (HSCs) is described. The isolation procedure is essentially based on magnetic enrichment of c-Kit+ cells and cell sorting to purify HSCs for cellular and molecular studies.

We demonstrate how to identify and isolate 6 subsets of myeloid progenitors from murine bone marrow using a combination of magnetic and fluorescence sorting (MACS and FACS). This protocol can be used for in vitro culture assays (methylcellulose or liquid cultures), in vivo adoptive transfer experiments, and RNA/protein analyses.

The fate of an individual embryonic cell can be influenced by inherited molecules and/or by signals from neighboring cells. Utilizing fate maps of the cleavage stage Xenopus embryo, single blastomeres can be identified for culture in isolation to assess the contributions of inherited molecules versus cell-cell interactions.
