Spot Forming Units

Spot forming units (SFUs) are a quantitative measure of individual cells that release a detectable molecule, most commonly a cytokine or antibody, in immunological assays. In an ELISpot assay, secreted products bind capture antibodies on a membrane, are detected with labeled antibodies, and form localized spots, with each spot generally representing one secreting cell. Researchers report SFUs relative to a defined number of cells to assess antigen-specific T- or B-cell responses, compare immune activation, and evaluate infection, vaccination, or immunotherapy. SFU measurements provide a sensitive, functional readout of cellular immunity while requiring relatively small samples.

Here, we describe a novel flow cytometric method for prospective isolation of early burst-forming unit erythroid (BFU-e) and colony-forming unit erythroid (CFU-e) progenitors directly from fresh mouse bone marrow and spleen. This protocol, developed based on single-cell transcriptomic data, is the first to isolate all the tissue's erythroid progenitors with high purity.
