Kidney Injury Model

A kidney injury model is an experimental system that reproduces structural or functional damage to the kidney, enabling researchers to investigate disease mechanisms and potential treatments. Models may induce injury through ischemia-reperfusion, nephrotoxic compounds, urinary obstruction, or controlled damage in cultured renal cells, allowing analysis of tubular dysfunction, inflammation, oxidative stress, and tissue repair. In medicine, kidney injury models support research on acute kidney injury and chronic renal disease, help evaluate biomarkers and therapeutic strategies, and provide controlled settings for studying how damaged nephrons recover or progress toward fibrosis. Their findings can guide preclinical studies and improve understanding of patient outcomes.

Ischemia-reperfusion Model of Acute Kidney Injury and Post Injury Fibrosis in Mice

We describe models of moderate and severe ischemia-reperfusion-induced kidney injury in which the mice undergo unilateral renal pedicle clamping followed by simultaneous or delayed contralateral nephrectomy, respectively. These models consistently give rise to renal dysfunction and post-injury fibrosis, but injury severity and survival are dependent on mouse background, age and surgical equipment.

Renal injuries incurred from nephrotoxins, which include drugs ranging from antibiotics to chemotherapeutics, can result in complex disorders whose pathogenesis remains incompletely understood. This protocol demonstrates how zebrafish can be used for disease modeling of these conditions, which can be applied to the identification of renoprotective measures.

This work presents a new in vivo model of segmental kidney injury using kidney GFP transgenic zebrafish. The model allows for the induction of the targeted ablation of kidney epithelial cells to show the cellular mechanisms of nephron injury and repair.

Intravenous Microinjections of Zebrafish Larvae to Study Acute Kidney Injury

We describe a technique of microinjecting the aminoglycoside, gentamicin, into 2 days post-fetilization (dpf) zebrafish larvae to induce acute kidney injury (AKI). We also describe a method for whole mount immunohistochemistry, plastic embedding and sectioning of zebrafish larvae to visualize the AKI mediated damage.
