Aav Genome Copy

AAV genome copy is a measurement of the number of adeno-associated virus (AAV) vector genomes present in a sample, providing a key indicator of vector dose and preparation quality. It is typically determined by extracting nucleic acids and using quantitative PCR or digital PCR to amplify a defined sequence, with the resulting signal compared with standards or partition-based counting to estimate genome copies. In neuroscience, this measurement helps researchers normalize AAV doses delivered to neuronal cultures or brain tissue, compare transduction experiments, and assess vector distribution and persistence. Genome-copy data support reproducible studies of neural circuits, gene expression, and therapeutic delivery while distinguishing vector quantity from functional expression.

Isolation and Genome Analysis of Single Virions using 'Single Virus Genomics'

Single Virus Genomics (SVG) is a method to isolate and amplify the genomes of single virons. Viral suspensions of a mixed assemblage are sorted using flow cytometry onto a microscope slide with discrete wells containing agarose, thereby capturing the virion and reducing genome shearing during downstream processing. Whole genome amplification is achieved using multiple displacement amplification (MDA) resulting in genomic material that is suitable for sequencing.

In this video, we demonstrate an in vitro colorimetric assay to detect the presence of neutralizing antibodies against alkaline phosphatase-expressing recombinant adeno-associated virus serotype 6 (rAAV6) vectors in sheep serum samples. The assay utilizes the reaction between the AAV encoding a human placental alkaline phosphatase gene and its substrate to generate an insoluble purple product.
