Calcium Dye Loading

Calcium dye loading is the process of introducing calcium-sensitive fluorescent indicators into living cells to monitor changes in intracellular calcium, a key signal in neuronal communication. Membrane-permeable dyes, often provided as acetoxymethyl ester forms, enter cells and are cleaved by intracellular esterases, trapping the indicator inside; calcium binding then alters its fluorescence according to the dye’s properties. In neuroscience, researchers use loaded neurons or brain tissue to visualize activity through calcium imaging, linking fluorescence changes to patterns of excitation, synaptic signaling, and network dynamics. Careful control of loading conditions helps preserve cell health and produce reliable measurements.

Targeted-esterase induced dye loading (TED) supports the analysis of intracellular calcium store dynamics by fluorescence imaging. The method bases on targeting of a recombinant Carboxylesterase to the endoplasmic reticulum (ER), where it improves the local unmasking of synthetic low-affinity Ca2+ indicator dyes in the ER lumen.

Here, we describe a method for loading a calcium-sensitive dye through the frog nerve stump into the nerve endings. We also present a protocol for the recording and analysis of fast calcium transients in the peripheral nerve endings.

Assessing T-cell Receptor-Induced Calcium Influx Using a Calcium Indicator Dye

This video analyzes T-cell receptor-induced calcium influx in T-cells. Anti-CD3 antibodies activate the T-cell signaling pathways, leading to an increase in intracellular calcium ions that bind with Indo-1 dye. In flow cytometry, a spectrum shift from the calcium-free state to the calcium-bound state correlates with T-cell receptor-induced calcium influx.

Calcium is a ubiquitous messenger in the nervous system, essential for triggering neurotransmitter release and changes in synaptic strength. Here we demonstrate a technique for loading Ca2+-indicators into Drosophila nerve terminals. We also demonstrate fabrication of the required apparatus and emphasize points critical for the technique's success.
