Primary Mouse Neurons

Primary mouse neurons are nerve cells isolated directly from mouse brain or spinal tissue and maintained in culture, providing an experimentally accessible model of neuronal structure and function. After tissue dissociation, cells attach to a culture substrate and extend neurites under defined conditions, forming networks that support electrical signaling and synaptic communication. These cultures enable researchers to examine neuronal development, axon and dendrite growth, gene expression, and responses to drugs or injury in neuroscience research. Because they preserve many features of differentiated neurons, primary mouse neurons help connect cellular mechanisms with nervous-system function while complementing studies using animal models and engineered cell systems.

Dopaminergic neurons play a vital regulatory role in the brain. Their loss is associated with Parkinson's disease. In this video, we show how to generate primary cultures of central dopaminergic neurons from embryonic mouse mesencephalon. Such cultures are useful to study the extreme vulnerability of these neurons to various stresses.

Modeling Neuronal Injury in Mouse Primary Cerebellar Granule Neurons

This video demonstrates the generation of a neuronal injury model in mouse primary cerebellar granule neurons. It outlines the steps involved in inducing neuronal injury using NMDA, an excitatory neurotransmitter receptor agonist, and glycine, a co-agonist. NMDA and glycine bind to specific neuronal receptors, causing their overstimulation and resulting in a massive intracellular calcium influx, which leads to neuronal cell damage and death.

This protocol describes a simple method for isolating and culturing primary mouse cerebral granule neurons (CGNs) from 6-7 day old pups, efficient transduction of CGNs for loss and gain of function studies, and modelling NMDA-induced neuronal excitotoxicity, low-potassium-induced cell death, DNA-damage, and oxidative stress using the same culture model.

Isolation and Culture of Primary Embryonic Mouse Midbrain Dopamine Neurons

2025

The video demonstrates the process of isolating and culturing dopamine neurons from the midbrain of mouse embryos. The isolated ventral midbrain floors are dissociated using enzymes. The resulting dopamine neurons are harvested and transferred into micro-islands inside multi-well plates. The cells are incubated with a dopamine neuron medium to maintain the cells at a high density inside the micro-islands.

Primary Neuronal Cultures

2023

The complexity of the brain often requires neuroscientists to use a simpler system for experimental manipulations and observations. One powerful approach is to generate a primary culture by dissecting nervous system tissue, dissociating it into single cells, and growing those cells in vitro. Primary cultures make neurons and glia easily accessible to the experimental tools required for techniques like genetic manipulation and time-lapse imaging. Furthermore, these cultures represent a highly...
