Rare-lacz Reporter Cells

Rare-lacz reporter cells are cells containing a lacZ reporter gene that enables researchers to identify and study infrequent cellular events, making them useful for detecting sparse gene expression or cell populations in neuroscience. The lacZ gene encodes β-galactosidase, which converts substrates such as X-gal into a visible blue product, allowing labeled cells to be distinguished by histochemical or microscopic analysis. This approach supports investigations of neuronal development, cellular lineage, circuit organization, and disease-related changes when only a small number of cells express the reporter. By converting molecular activity into a stable visual signal, rare-lacz reporters help connect gene regulation with cellular location and function.

2025

This video demonstrates a technique for assessing retinoic acid activity in mouse embryos. Upon isolating cells from the mouse embryos, the cells are co-cultured with retinoic acid (RA) reporter cells. Embryonic cells produce RA, which induces beta-galactosidase expression in reporter cells, indicating RA activity in the embryonic cells.

Flow cytometry is a powerful tool allowing for the isolation and study of specific cell populations. This protocol describes steps for isolating LacZ-expressing cells from cochlear tissues from neonatal transgenic mice. Dissociated cochlear cells were labeled using fluorescent-conjugated substrates of β-galactosidase prior to separation via flow cytometry.
