Viral Protein Expression

Viral protein expression is the production of proteins encoded by viral genetic material within infected or engineered host cells, a process central to understanding viral biology. After viral nucleic acids enter a cell, they use host transcription and translation machinery, together with virus-specific regulatory signals, to produce proteins that support genome replication, particle assembly, and release. Researchers use viral protein expression to study infection mechanisms, identify interactions between viral and cellular components, and evaluate antiviral strategies. It also supports the development of recombinant vaccine antigens, diagnostic assays, and experimental systems for investigating viral protein structure and function.

We describe the usage of a fluorescent reporter vaccinia virus that enables real-time measurement of viral infectivity and gene expression through the stage-specific expression of spectrally distinct reporter fluorophores. We detail a plate-based method for accurately identifying the stage at which virus replication is affected in response to small molecule inhibition.
