Tumor Cell Interaction

Tumor cell interaction refers to the physical contact and molecular communication between cancer cells and surrounding cells, tissues, and extracellular matrix, processes that shape tumor development and progression. Through cell adhesion, receptor-mediated signaling, secreted factors, and extracellular vesicles, tumor cells can alter stromal and immune cell behavior while receiving signals that promote proliferation, invasion, survival, or immune evasion. Studying these interactions helps clarify how the tumor microenvironment supports metastasis and treatment resistance. Experimental models of tumor cell interaction also inform biomarker discovery, therapeutic design, and strategies that target cancer cells together with the supportive cells and signals around them.

Three-dimensional Co-culture Model for Tumor-stromal Interaction

Here we present a protocol to co-culture in three-dimensions, which is useful for investigating multicellular interactions and extracellular matrix-dependent modulation of cancer cell behavior. In this experimental model, cancer cells are cultured on collagen gels embedded with human cancer-associated fibroblasts.

We have developed 3D coculture models for live-cell imaging in real-time of interactions among breast tumor cells and other cells in their microenvironment that impact progression to an invasive phenotype. These models can serve as preclinical screens for drugs to target paracrine-induced proteolytic, chemokine/cytokine and kinase pathways implicated in invasiveness.

Given the limited xenograft models available to study interactions between human CART and myeloid cells, we established in vitro and in vivo models to understand the impacts of human macrophages on CART cells. Findings can potentially be generalized to evaluate macrophage roles in the tumor microenvironment and test macrophage-targeted immunotherapies.
