Organoid Transfection

Organoid transfection is the introduction of DNA, RNA, or genome-editing reagents into three-dimensional cell cultures that model aspects of human tissues. In this process, lipid-based carriers or electroporation help nucleic acids cross cell membranes, enabling transient expression or, when paired with editing systems, targeted genetic modification in organoid cells. In genetics research, the method supports gene-function studies, disease modeling, lineage tracing, and evaluation of regulatory elements in a tissue-like context. By enabling controlled manipulation of organoid genomes while preserving cellular organization, organoid transfection links molecular changes to tissue-level phenotypes and advances studies of development, disease mechanisms, and therapeutic responses.

Transgenic mice or viral vectors have been used to increase protein expression within the lung. However, these techniques are time-consuming, technically challenging and have off-target effects that can confound results. Our protein transfection protocol uses a lipid based transfection reagent and an ultrafine microsprayer to uniformly deliver active protein to lung cells.
