Myelin Oligodendrocyte Glycoprotein

Myelin oligodendrocyte glycoprotein (MOG) is a surface protein expressed by oligodendrocytes, the cells that produce myelin in the central nervous system. Its extracellular immunoglobulin-like domain is exposed at the outer myelin surface, making MOG accessible to antibodies and immune cells and allowing it to serve as an antigen in neuroinflammatory responses. In immunology, MOG is used to study antibody-mediated demyelination and immune tolerance, including experimental models of multiple sclerosis-like disease. Detecting MOG-specific antibodies also supports the evaluation and classification of MOG antibody-associated disease, improving understanding of autoimmune injury to the brain, spinal cord, and optic nerves.

We describe a simple protocol using only basic lab equipment to generate and purify large quantities of a fusion protein that contains mouse Myelin Oligodendrocyte Glycoprotein. This protein can be used to induce experimental autoimmune encephalomyelitis driven by both T and B cells.

This article describes methods to derive enriched populations of murine oligodendrocyte precursor cells (OPCs) in primary culture, which differentiate to produce mature oligodendrocytes (OLs). In addition, this report describes techniques to produce murine myelinating co-cultures by seeding mouse OPCs onto a neurite bed of mouse dorsal root ganglion neurons (DRGNs).

The video demonstrates a protocol for differentiating oligodendrocyte precursor cells (OPCs) into oligodendrocytes and inducing myelination in a co-culture with dorsal root ganglion (DRG) neurons. The OPCs are transduced with lentiviral vectors encoding a signaling protein that regulates myelination. The transduced OPCs are co-cultured with DRG neurons to assess the differentiation into oligodendrocytes and the signaling protein-induced myelination.

Experimental autoimmune encephalomyelitis (EAE) is an established animal model of multiple sclerosis. C57BL/6 mice are immunized with myelin oligodendrocyte glycoprotein (MOG) peptide 35-55 (MOG35-55), resulting in an ascending flaccid paralysis caused by autoreactive immune cells in the central nervous system. Protocols for disease induction and monitoring will be discussed.
