Fluorescent Chimeric Mice

Fluorescent chimeric mice are animal models that combine cells or tissues from genetically distinct sources with fluorescent labeling, enabling researchers to visualize biological events in living organisms. Typically, cells engineered to express fluorescent proteins, such as green fluorescent protein, are introduced into developing embryos or transplanted into recipient mice, where microscopy tracks their location, survival, and behavior. In medicine, these models support studies of cancer growth, immune-cell trafficking, tissue regeneration, and cell transplantation. By linking cell identity with spatial and temporal imaging, fluorescent chimeric mice help evaluate disease mechanisms, treatment responses, and the integration of therapeutic cells in complex physiological environments.

We describe a protocol whereby busulfan conditioning permits the bone marrow of a recipient mouse to be replaced with bone marrow cells from donor mice ubiquitously expressing green fluorescent protein, in the absence of irradiation. This technique is useful to study bone marrow cell accumulation in the central nervous system.
