Peritoneal Surface Staining

Peritoneal surface staining is a medical visualization technique that uses tissue-reactive dyes or histological markers to make features of the peritoneum more apparent during examination or analysis. The applied stain interacts differently with normal mesothelium, damaged tissue, inflammatory areas, or abnormal cellular deposits, creating contrast that helps distinguish surface changes. In clinical medicine and pathology, this approach can support the assessment of peritoneal disease, guide targeted sampling, and clarify the distribution of lesions. It also provides a practical way to correlate visible surface patterns with microscopic findings and disease mechanisms.

The peritoneal cavity in mammals contains different immune cell populations crucial for innate immune responses. An efficient isolation method is required for biochemical and functional analyses of these cells. Here we provide a comprehensive method for the isolation of peritoneal cavity cells in the mouse.

Quantitation of Intra-peritoneal Ovarian Cancer Metastasis

Ovarian cancer metastasis is characterized by numerous diffuse intra-peritoneal lesions, such that accurate visual quantitation of tumor burden is challenging. Herein we describe a method for in situ and ex vivo quantitation of metastatic tumor burden using red fluorescent protein (RFP)-labeled tumor cells and optical imaging.

In this video, we demonstrate the development of an in vitro three-dimensional model of the lining of the peritoneal cavity. This organotypic culture can help to investigate ovarian cancer cell adhesion, invasion, and proliferation.

Peritoneal Low-density Neutrophil Isolation: A Technique to Obtain Low-density Neutrophils from Peritoneal Lavage Fluid Using Magnetic Activated Cell Sorting

In this video, we describe the procedure for isolating low-density neutrophils, or LDNs, from the peritoneal lavage fluid of a postoperative human patient. The LDNs thus obtained can be used in further downstream experiments to understand their role in tumor growth and progression.
