Neuronal Analysis

Neuronal analysis is the systematic study of neuron structure, activity, connectivity, and function, providing essential insight into how nervous systems process information. Researchers combine methods such as microscopy, electrophysiology, calcium imaging, and molecular labeling to examine features ranging from cellular morphology to membrane potential, action potentials, synaptic signaling, and network activity. In neuroscience, these approaches help characterize neuronal circuits, compare healthy and diseased tissue, and relate cellular events to behavior and cognition. Neuronal analysis also supports research on neurodevelopment, neurodegeneration, brain injury, and therapeutic strategies by revealing how changes in individual neurons influence broader nervous system function.

Cell Subtype-specific Analysis of Neuronal Membrane Proteasome in Somatosensory Neurons

The neuronal membrane proteasome (NMP) was recently identified in a subset of somatosensory neurons as a key regulator of touch, pain, and itch perception. This article presents a robust workflow for analyzing cell-type-specific NMP expression and function using cell sorting, transcriptome profiling, and culture-based immunofluorescent analyses.

A Neuronal and Astrocyte Co-Culture Assay for High Content Analysis of Neurotoxicity

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.

Numerous recent studies have identified mutations in synaptic proteins associated with brain pathologies. Primary cultured cortical neurons offer great flexibility in examining the effects of these disease-associated proteins on dendritic spine morphology and motility.

Automated Sholl Analysis of Digitized Neuronal Morphology at Multiple Scales

We have developed a computer program to analyze neuronal morphology. In combination with two existing open source analysis tools, our program performs Sholl analysis and determines the number of neurites, branch points, and neurite tips. The analyses are performed so that local changes in neurite morphology can be observed.

We describe how to use laser capture microdissection (LCM) to obtain enriched populations of hippocampal neurons or single neurons from frozen sections of the injured rat brain for subsequent gene expression analysis using quantitative real time PCR and/or whole-genome microarrays.
