Primary Microglia

Primary microglia are microglial cells isolated directly from central nervous system tissue and maintained in culture to study the brain’s resident immune cells. They respond to environmental cues through innate immune receptors, changing morphology and gene expression while releasing inflammatory mediators, engulfing cellular debris, and supporting tissue homeostasis. Researchers use primary microglia to investigate neuroinflammation, synaptic remodeling, neurodegenerative disease, infection, and responses to potential therapeutics. Because these cells preserve many features of their tissue origin, primary microglia provide a valuable experimental model for examining how immune signaling influences neuronal health and disease in neuroscience.

We outline methods for the efficient and quick isolation/culture of viable microglia from the neonatal cerebral cortex and adult spinal cord. The dissection and plating of cortical microglia can be accomplished within 90 minutes, with the subsequent microglial harvest taking place ~ 10 days following the initial dissection.
