Small-scale Aav Production

Small-scale AAV production is the laboratory-scale generation of recombinant adeno-associated virus vectors for delivering genetic material into target cells, a process central to neuroscience research and gene therapy development. Typically, producer cells are transiently transfected with plasmids encoding the therapeutic or experimental genome, viral replication and capsid proteins, and helper functions; assembled particles are then harvested, purified, and assessed for genome titer and quality. These vectors can introduce genes into neurons and glial cells, enabling studies of neural circuits, disease mechanisms, and therapeutic strategies. Small-scale preparation supports early construct testing, serotype comparison, and optimization before larger-scale production.

Synthetic Spider Silk Production on a Laboratory Scale

Despite the outstanding mechanical and biochemical properties of spider silks, this material cannot be harvested in large quantities by conventional means. Here we describe an efficient strategy to spin artificial spider silk fibers, which is an important process for investigators studying spider silk production and their use as next-generation biomaterials.

We demonstrate the basic technique to molecularly engineer and evolve synthetic Adeno-associated viral (AAV) gene therapy vectors via DNA family shuffling. Moreover, we provide general guidelines and representative examples for selection and analysis of individual chimeric capsids with enhanced properties on target cells in culture or in mice.
