Bacterial Cell Isolation

Bacterial cell isolation is a laboratory technique used to separate bacteria from mixed samples and obtain a population suitable for controlled study. It typically relies on dilution and spatial separation, followed by growth on solid culture media under appropriate conditions so individual cells or small groups form distinct colonies; selective or differential media can help distinguish target organisms. In biochemistry, isolated cultures support reliable analysis of cellular components, enzyme activity, metabolism, and genetic traits. The resulting pure or enriched cultures improve experimental reproducibility and enable bacterial identification, characterization, and investigation of microbial functions.

Isolation and Identification of Bacterial Strains from Skin of Terrestrial Amphibians

2025

This is a method for sampling and isolating culturable bacteria from the cutaneous microbiota of European plethodontid salamanders (Speleomantes genus). Here, we present a protocol for sampling salamander skin with swabs and processing them using culturable approaches. We describe sampling, isolation, and establishment of axenic culture with bacterial strain characterization.

The bacterial cell wall is composed of peptidoglycan, a macromolecular network of sugar strands crosslinked by peptides. Ultra Performance Liquid Chromatography provides high resolution and throughput for novel discoveries of peptidoglycan composition. We present a procedure for the isolation of cell walls (sacculi) and their subsequent preparation for analysis via UPLC.

Isolation, Propagation, and Identification of Bacterial Species with Hydrocarbon Metabolizing Properties from Aquatic Habitats

2021

We present the process of isolating, propagating, and characterizing hydrocarbon-degrading bacteria from aquatic habitats. The protocol outlines bacterial isolation, identification by the 16S rRNA method, and testing of their hydrocarbon-degrading potential. This article would help researchers in characterizing microbial biodiversity in environmental samples, and specifically screen for microbes with bioremediation potential.
