Pancreatic Lineage Induction

Pancreatic lineage induction is the directed differentiation of pluripotent stem cells or other progenitors into pancreatic cell types, providing a foundation for studying development and engineering replacement tissues. The process recreates stages of embryonic development by sequentially modulating signaling pathways that establish definitive endoderm, posterior foregut, pancreatic progenitors, and specialized endocrine or exocrine cells. In bioengineering, these induced cells support organoid and tissue fabrication, disease modeling, drug screening, and research on diabetes and pancreatic disorders. Improved control over cell identity, maturation, and organization may advance cell-based therapies and the development of functional pancreatic tissues.

This protocol presents a comparison between two different induction protocols for differentiating human dental pulp stem cells (hDPSCs) toward pancreatic lineages in vitro: the integrative protocol and the non-integrative protocol. The integrative protocol generates more insulin producing cells (IPCs).

Protocols to generate hPSC mutant lines using the iCRISPR platform and to differentiate hPSCs into glucose-responsive β-like cells are described. Combining genome editing technology with hPSC-directed differentiation provides a powerful platform for the systematic analysis of the role of lineage determinants in human development and disease progression.

Isolation of Stem Cells from Human Pancreatic Cancer Xenografts

Cancer stem cells (CSCs) have been identified in a number of malignancies. In this protocol we describe a flow cytometric method utilizing aldehyde dehydrogenase activity and CD44 and CD24 expression to isolate CSCs from human pancreatic adenocarcinoma xenografts. These viable cells can then be used in functional and analytical studies.

Here, we present a protocol for a mosaic labeling technique that permits the visualization of neurons derived from a common progenitor cell in two distinct colors. This facilitates neural lineage analysis with the capability of birth-dating individual neurons and studying gene function in the same neurons of different individuals.
