Environmental Genomic Libraries

Environmental genomic libraries are collections of DNA fragments recovered from environmental samples, such as soil, water, or sediment, that represent the genetic material of entire microbial communities. Researchers extract and fragment environmental DNA, insert the fragments into cloning vectors or sequence them directly, and analyze the resulting library to identify genes, metabolic pathways, and organisms without cultivating each microbe. These libraries support metagenomic studies of biodiversity, nutrient cycling, and microbial adaptation, while also enabling the discovery of novel enzymes, antibiotics, and other molecules with applications in biotechnology, environmental monitoring, and medicine.

Large Insert Environmental Genomic Library Production

Construction of a fosmid library with environmental genomic DNA isolated from the vertical depth continuum of a seasonally hypoxic fjord is described. The resulting clone library is picked into 384-well plates and archived for downstream sequencing and functional screening by the application of an automated colony picking system.

This manuscript describes an automated gel size selection approach for purifying DNA fragments for next-generation sequencing. The Ranger Technology provides complete automation of the entire process of agarose gel loading, electrophoretic analysis, and recovery of targeted DNA fragments allowing for high-throughput and high quality next-generation sequencing libraries.

Isolation and Genome Analysis of Single Virions using 'Single Virus Genomics'

Single Virus Genomics (SVG) is a method to isolate and amplify the genomes of single virons. Viral suspensions of a mixed assemblage are sorted using flow cytometry onto a microscope slide with discrete wells containing agarose, thereby capturing the virion and reducing genome shearing during downstream processing. Whole genome amplification is achieved using multiple displacement amplification (MDA) resulting in genomic material that is suitable for sequencing.
