F-actin Biosensor

An F-actin biosensor is a molecular probe that detects and visualizes filamentous actin, the polymerized form of actin that supports cell shape, movement, and force generation. Typically, a fluorescent label is linked to an actin-binding component that associates with F-actin, causing the probe to concentrate along actin filaments and reveal their organization and remodeling through microscopy. These biosensors help researchers monitor cytoskeletal dynamics during cell migration, adhesion, division, and shape changes, providing insight into how cells coordinate mechanical behavior with signaling. They are valuable tools in cell biology, developmental biology, and disease research involving altered cytoskeletal regulation.

Filamentous actin (F-actin) plays an important role in spinogenesis, synaptic plasticity, and synaptic stability. Quantification of F-actin puncta is therefore a useful tool to study the integrity of synaptic structures. This protocol describes the procedures of quantifying F-actin puncta labeled with Phalloidin in low-density primary cortical neuronal cultures.

In Vivo Biosensor Tracks Non-apoptotic Caspase Activity in Drosophila

To detect healthy cells in whole animals that contain low levels of caspase activity, the highly sensitive biosensor designated CaspaseTracker was generated for Drosophila. Caspase-dependent biosensor activity is detected in long-lived healthy cells throughout the internal organs of adult animals reared under optimized conditions in the absence of death stimuli.

Microscopic imaging of live endothelial cells expressing GFP-actin allows characterization of dynamic changes in cytoskeletal structures. Unlike techniques that use fixed specimens, this method provides a detailed assessment of temporal changes in the actin cytoskeleton in the same cells before, during, and after various physical, pharmacological, or inflammatory stimuli.
