Spirochetes Pleomorphic Bacteria

Spirochetes are slender, spiral-shaped bacteria that move through liquid or viscous environments, while pleomorphic bacteria can adopt variable shapes and sizes under different conditions. In spirochetes, periplasmic flagella, also called endoflagella, lie between the cell membrane and outer sheath; their rotation bends and twists the cell into a corkscrew form that drives motility. These contrasting morphological traits help biologists classify bacteria, interpret microscopic observations, and understand how cell structure supports movement and survival. Studying spirochetes and bacterial pleomorphism also informs research on host colonization, disease mechanisms, environmental adaptation, and the relationships between bacterial form and function.

Source: Eggers, C. H. Phage-Mediated Genetic Manipulation of the Lyme Disease Spirochete Borrelia burgdorferi. J. Vis. Exp. (2022).This video demonstrates the transduction of genetically modified Borrelia spirochetes using polyethylene glycol–precipitated, non-infectious bacteriophages carrying a kanamycin-resistance plasmid. After incubation with the bacteria, the culture is plated on antibiotic-supplemented medium. Only cells that receive the plasmid survive and form colonies. These colonies...

The ability of bacteriophage to move DNA between bacterial cells makes them effective tools for the genetic manipulation of their bacterial hosts. Presented here is a methodology for inducing, recovering, and using φBB-1, a bacteriophage of Borrelia burgdorferi, to transduce heterologous DNA between different strains of the Lyme disease spirochete.

Detection of Bacteria Using Fluorogenic DNAzymes

We have recently reported a novel approach for generating fluorogenic DNAzyme probes that can be applied to set up a simple, "mix-and-read" fluorescent assay for bacterial detection. These special DNA probes catalyze the cleavage of a chromophore-modified DNA-RNA chimeric substrate in the presence of crude extracellular mixture (CEM) produced by a specific bacterium, thereby translating bacterial detection into fluorescence signal generation. In this report we will describe key experimental...
