Iterative Experimentation

Iterative experimentation is a systematic process of repeating experiments, evaluating results, and refining hypotheses or procedures to improve scientific understanding and outcomes. In chemistry, researchers vary selected conditions such as reactant ratios, temperature, solvent, or reaction time, then use observations and analytical data to guide the next experimental cycle. This feedback-driven approach supports reaction optimization, synthesis development, and the investigation of structure–property relationships in chemical systems. By turning unexpected results into actionable information, iterative experimentation helps chemists identify influential variables, reduce inefficient trial and error, and develop more reliable methods for research and industrial applications.

This article describes a method to obtain a three-dimensional (3D) structure of helically assembled molecules using cryo-electron microscopy. In this protocol, we use HIV-1 capsid assemblies to illustrate the detailed 3D reconstruction procedure for achieving a density map by the iterative helical real-space reconstruction method.
