Swine Ischemia Model

The Swine Ischemia Model is an in vivo experimental system that uses pigs to investigate how reduced blood flow affects tissues and organs, providing a physiologically relevant platform for studying ischemic injury. Researchers induce or monitor a defined interruption of perfusion, which limits oxygen and glucose delivery, alters cellular metabolism, and can trigger tissue damage during ischemia and subsequent reperfusion. In developmental biology, this model helps examine how impaired circulation influences tissue growth, vascular development, cellular survival, and recovery. Its anatomical and physiological similarities to humans also support evaluation of disease mechanisms, protective interventions, and regenerative strategies.

The purpose of this protocol is to demonstrate the placement of a delayed constricting device (an ameroid constrictor) around a coronary artery in a swine model. This device creates an ischemic area of the heart that is useful for studying new diagnostic imaging techniques and new methods of treatment.

Here we describe the detailed procedure of intestinal ischemia-reperfusion in mice which results in reproducible injury without mortality to encourage the standardization of this technique across the field. This model of intestinal ischemia-reperfusion injury can be utilized to study the cellular and molecular mechanisms of injury and regeneration.
