Flat Mount Choroid Quantification

Flat mount choroid quantification is an imaging-based method for measuring the structure and extent of the choroidal vascular network after the tissue is dissected, flattened, and labeled for visualization. Researchers typically remove the retina, make radial incisions in the choroid-sclera complex, mount the tissue as a flat preparation, and use microscopy with image-analysis software to quantify parameters such as vessel area, density, branching, or lesion size. In medicine, this technique supports studies of ocular vascular biology and diseases involving abnormal choroidal neovascularization, including age-related macular degeneration, while enabling comparison of treatment effects and disease progression.

The branchial skeleton, including gill rakers, pharyngeal teeth, and branchial bones, serves as the primary site of food processing in most fish. Here we describe a protocol to dissect and flat-mount this internal skeleton in threespine sticklebacks. This method is also applicable to a variety of other fish species.

The zebrafish embryo is an excellent model for developmental biology research. During embryogenesis, zebrafish develop with a yolk mass, which presents three-dimensional challenges for sample observation and analysis. This protocol describes how to create two-dimensional flat mount preparations of whole mount in situ (WISH) stained zebrafish embryo specimens.

Retinal prostheses have the ability to generate visual perceptions. To advance the development of new prostheses, ex vivo methods are needed to test devices before implantation. This article provides a comprehensive protocol for studying calcium activity in the retinal ganglion cell layer when subjected to electrical stimulation.

Establishing a Whole-Cell Patch Clamp for Electrophysiological Recording from a Flat-Mount Mouse Retina

2025

This video demonstrates a whole-cell patch-clamp technique for electrophysiological recording from a flat-mount mouse retina. A thin glass pipette with an electrode is used to approach starburst amacrine cells (SACs) within the retina. After forming a seal, suction is applied to rupture the membrane and form the whole-cell patch-clamp configuration. Finally, the flow of ions through the SAC synaptic receptors is measured using the configuration.
