Laser-assisted Anastomosis

Laser-assisted anastomosis is a microsurgical technique that uses focused laser energy to join tubular tissues, especially blood vessels, with high spatial precision. The laser produces localized photothermal effects that denature and fuse collagen at apposed tissue edges, creating a seal while reducing the need for conventional sutures. Surgeons can apply this approach in vascular reconstruction and other delicate procedures where minimizing foreign material, tissue handling, and lumen narrowing is important. By supporting controlled tissue repair and potentially shortening technically demanding anastomoses, laser-assisted anastomosis contributes to advances in microsurgery and reconstructive medicine.

Here we present a protocol for laser-assisted microdissection of specific plant cell types for transcriptional profiling. While the protocol is suitable for different species and cell types, the focus is on highly inaccessible cells of the female germline important for sexual and apomictic reproduction in the crucifer genus Boechera.
