Autonomic Innervation

Autonomic innervation is the neural supply of smooth muscle, cardiac muscle, glands, and internal organs by the autonomic nervous system, enabling largely involuntary control of homeostasis. Preganglionic neurons in the central nervous system transmit signals to peripheral ganglia, where postganglionic neurons release neurotransmitters that adjust organ activity through sympathetic, parasympathetic, and enteric pathways. These pathways regulate heart rate, blood pressure, digestion, pupil diameter, and secretion through opposing or coordinated effects. Mapping autonomic innervation helps researchers understand visceral reflexes, drug actions, and disorders involving cardiovascular, gastrointestinal, and nervous-system function.

Standardized, comprehensive and fully quantitative testing of autonomic functions is described. The autonomic tests consist of evaluation of all three major autonomic domains including cardiovagal, adrenergic and sudomotor. The severity and distribution of dysautonomia is quantitated using Composite Autonomic Severity Scores.

Compound muscle action potential recording quantitatively assesses functional diaphragm innervation by phrenic motor neurons. Whole-mount diaphragm immunohistochemistry assesses morphological innervation at individual neuromuscular junctions. The goal of this protocol is to demonstrate how these two powerful methodologies can be used in various rodent models of spinal cord disease.

A Method to Target and Isolate Airway-innervating Sensory Neurons in Mice

Organ specific sensory neurons are difficult to identify. Fast Blue tracing is used to identify nodose neurons innervating the airways for cell sorting. Sorted nodose neurons are used to extract high quality ribonucleic acid (RNA) for sequencing. Using this protocol, gene expression of airway specific neurons is determined.
