Parasympathetic Nerve Transection

Parasympathetic nerve transection is the deliberate surgical severing of a parasympathetic nerve to interrupt autonomic communication between the central nervous system and a target organ. By cutting the nerve, researchers stop action-potential transmission and cholinergic efferent signaling, producing denervation whose physiological effects depend on the affected pathway and organ. In neuroscience, this method helps isolate parasympathetic contributions to functions such as heart rate, glandular secretion, gastrointestinal motility, and smooth-muscle activity. Comparing intact and denervated animals or tissues can clarify autonomic control mechanisms, reveal compensatory changes, and support studies of neural injury and functional recovery.

Optic Nerve transection is a widely used model of adult CNS injury. Ninety percent of retinal ganglion cells (RGCs) whose axons are completely transected (axotomy) die within 14 days after axotomy. This model is easily amenable to experimental manipulations and highly reproducible.

Although the peripheral nervous system (PNS) is capable of significant repair after injury, little is known about the cellular and molecular mechanisms that govern this phenomenon. Using live, transgenic zebrafish and a reproducible nerve transection assay, we can study dynamic glial cell behaviors during nerve degeneration and regeneration.
